Biotinidase is a ubiquitous mammalian enzyme that cleaves biotin from biocytin, making the free cofactor available to biotin-dependent enzymes such as carboxylases. However, biotinidase complicates applications such as antibody pretargeting with a biotin-streptavidin system because it attacks the targeting system’s biotin.
In 2006, D. Scott Wilbur, Donald K. Hamlin, and Ming-Kuan Chyan discovered a small biotin modification that resists biotinidase cleavage. Biotin binding to avidin or streptavidin remains unchanged.
